Frequently asked questions about GenPlates

What kind of samples can you load onto a GenPlate?

Currently we have validated sample application protocols for whole blood, blood products, purified DNA and clones. We are working on sample application protocols for buccal cell swabs and fixed tissue.

Are there any requirements for handling or preparation of samples before they are applied to the GenPlates?

Yes, GenVault has developed a set of guidelines that will allow users to maximize DNA recovery. Please see the Sample Application Protocol. If buffy coat, genomic DNA, or buccal wash/swabs are being applied, then one would need to prepare those materials before applying them to the GenPlate.

How are the samples actually preserved on the paper?

Moisture in the sample activates chemicals in the FTA elements that lyse cells, denature enzymes, inactivate pathogens and immobilize genomic DNA. FTA chemicals also protect DNA and RNA from light, free radicals, enzymes and pathogens during dry, room-temperature storage.

How much whole blood is applied to each well?

Each GenPlate element has a capacity of about 10ul of whole blood.

How much sample do I need to fill a whole GenPlate?

You need approximately 4 ml (cc) of whole blood or genomic DNA to fill an entire GenPlate. One 7ml tube will fill ~75-100 elements with buffy coat. Six 7ml tubes are needed to fill an entire GenPlate with buffy coat.

Do I have to fill the whole plate?

It is not necessary to fill an entire GenPlate.

What does FTA stand for?

Professor Leigh Burgoyne is the inventor of FTA technology. It arose many years ago from Leigh's need to collect nucleic acid samples (particularly blood) and safely transport and store these prior to analysis (by PCR or RFLP). Moreover, he wanted a convenient medium that would preserve the nucleic acid and could facilitate fast analysis of multiple samples using a robot. The idea for the trade mark 'FTA' arose after much 'brainstorming' for a short name that would stick in people's minds. FTA was chosen to mean Fast Technology for Analysis (of nucleic acids).

How long can I store my sample on FTA paper and still recover useful DNA?

DNA stored on FTA paper for 17 years has been successfully amplified and genotyped.

Will anticoagulants affect the quality of my DNA?

Anti-coagulants will not affect the quality or quantity of DNA recovered.

Will humidity affect the storage of my DNA?

Yes, high levels of humidity will affect the storage of your DNA over the long term. Damage can be prevented by keeping your GenPlates in their foil pouches until you are ready to use them. After spotting, keep GenPlates in a GenVault archive or in a desiccated container when storing.

How do I know how many samples are taken from a plate and how many are still there?

The number of elements used can be tracked with the software provided with each GenVault system. Elements in the Personal Archive are manually tracked, whereas elements in the Dynamic Archive are automatically tracked.
