            A robust chemistry for recovering high quality double-stranded DNA preserved in Genplates™, FTA Paper and Guthrie cards.  GenSolve enables efficient recovery of high quality DNA from biosamples such as blood, buffy coat and plasmid DNA that have been dried on FTA paper and Guthrie cards and banked at ambient temperature. GenSolve is a novel chemistry that ensures excellent recovery of high molecular weight, double-stranded DNA for use with leading downstream molecular analysis platforms. GenSolve utilizes fast and simple protocols that preserve DNA integrity and ensure efficient recoveries.

Recover DNA on genPlates

Figure 1A
The molecular weight of DNA recovered using GenSolve was examined by running 50ng of unamplified DNA directly on a 0.8% agarose gel stained with ethidium bromide. Human genomic DNA from Roche was used as a control.

 

Figure 1B
Analysis by QPCR confirms reliable amplification of recovered DNA. The reactions amplified a region in exon 3 of the human β-actin gene, which was detected by a FAM-dye-labeled β-actin TaqMan probe. Blue line – 5ng of GenSolve sample; Red line – 5ng of Roche genomic DNA used as a control; Yellow line – No template control

 

Recover intact plasmids from GenPlates spotted with glycerol stocks, DMSO stocks or bacterial cultures for use in PCR and transformation

Intact plasmid DNA successfully recovered after storage in GenPlate and recovery using GenSolve. Samples are 20ng BlueScript plasmid with EST insert run on 0.8% agarose gel stained with ethidium bromide.

Transformation Efficiency of Plasmid DNA following Storage on GenPlates. 1.0 μg of each purified plasmid (pCMV or pBluescript) was applied to GenPlates and stored for 30 days at room temperature. Plasmid DNA stored at -20°C was used as a control. Plasmids were recovered from GenPlates using GenSolve and used directly for transformation. Equivalent transformation efficiency of 100% was achieved for plasmids stored on GenPlates or at -20°C.
